چکیده

S ENSITIZATION of animals with allogeneic blood platelets induces accelerated rejection of allogeneic skin as well as renal grafts.2 In the latter instance the histologic lesions of the rejected graft were described as primarily vascular. These as well as earlier observations have suggested that an antigenic relationship exists between blood platelets and vascular endothelium. The experiments to be reported here were undertaken with the objective of directly testing this possibility. Repeated injections of antigen derived from allogeneic aortic endothelium sensitized dogs to allogeneic blood platelets. This was demonstrated by a sharp reduction in the survival of allogeneic platelets in the circulation of the sensitized recipients.

Different Stages of Platelet Adhesion to the Site of Vascular Injury

Platelet activation and adhesion to the site of vascular injury is a dynamic process comprising reversible and irreversible phases. Platelet adhesion typically occurs in a multi-step process similar to the selectin/integrin-mediated adhesion of neutrophils. This phenomenon is highly regulated and influenced by the cross-talk between platelets and injured endothelium. This cross-talk involves a ...

Platelet – Leukocyte Interactions: Multiple Links Between Inflammation, Blood Coagulation and Vascular Risk

The aim of this review is to summarize the contribution of platelets and leukocytes and their interactions in inflammation and blood coagulation and its possible relevance in the pathogenesis of thrombosis. There is some evidence of an association between infection/inflammation and thrombosis. This is likely a bidirectional relationship. The presence of a thrombus may serve as a nidus of infect...

Role of free radicals in the pathogenesis of acute chest syndrome in sickle cell disease

Acute chest syndrome (ACS) of sickle cell disease (SCD) is characterized pathologically by vaso-occlusive processes that result from abnormal interactions between sickle red blood cells (RBCs), white blood cells (WBCs) and/or platelets, and the vascular endothelium. One potential mechanism of vascular damage in ACS is by generation of oxygen-related molecules, such as superoxide (O2-), hydrogen...

Bench-to-bedside review: Platelets and active immune functions - new clues for immunopathology?

Platelets display a number of properties besides the crucial function of repairing damaged vascular endothelium and stopping bleeding; these are exploited to benefit patients receiving platelet component transfusions, which might categorize them as innate immune cells. For example, platelets specialize in pro-inflammatory activities, and can secrete a large number of molecules, many of which di...

Platelets play an essential role in separating the blood and lymphatic vasculatures during embryonic angiogenesis.

RATIONALE Several mutations that impair the development of blood lineages in the mouse also impair the formation of the lymphatic vasculature and its separation from the blood vasculature. However, the basis for these defects has remained unknown because the mutations characterized affect more than one blood lineage. OBJECTIVE We tested the hypothesis that megakaryocytes/platelets are require...
